Dihydrofolate Reductase of Streptococcus f aecium
نویسنده
چکیده
From a single amethopterin-resistant organism, Streptococcus faecium var. durans strain A, two different dihydrofolate reductases have been obtained as essentially homogeneous proteins in good yield. One of the reductases has a similar substrate specificity and turnover number (about 8000 moles per min per mole of enzyme) to the single reductase found in the amethopterin-sensitive strain of S. faecium var. durans, ATCC 8043, and has therefore been designated “wild type.” The other enzyme, which is distinguished by its ability to catalyze the reduction of folate, in addition to dihydrofolate, and by its lower turnover number (about 900 with dihydrofolate), has been designated “mutant type.” Since the wild type and mutant type reductases have sedimentation constants (sZO,buffer) of 2.58 S and 2.04 S, respectively, they are probably significantly different in molecular weight. Each exhibits a single pH optimum at pH 5.8 and is inactivated by urea. Neither is affected by methyhnercuric salts but the wild type reductase is inactivated by phenylmercuric acetate and P-mercuribenzoate. Monovalent cations increase the activity of the mutant type reductase but decrease that of the wild type reductase. It is suggested that the amethopterin resistance in vivo of strain A depends at least partly on the folate reductase activity of the mutant type reductase.

متن کاملFolate reductase and specific dihydrofolate reductase activities of the amethopterin-sensitive Streptococcus faecium var. durans.
Preliminary evidence is presented that indicates that the dihydrofolate reductase activity of amethopterin-sensitive Streptococcus faecium var. durans ATCC 8043 is separable into two dihydrofolate reductases, one of which also reduces folate.

Iclaprim, a novel dihydrofolate reductase inhibitor, inhibited 90% of the clinical isolates (MIC(90)) of Streptococcus pneumoniae (n = 785) collected by a national surveillance program at a concentration of 1 microg/ml. The MIC(90) for iclaprim was 7 doubling dilutions lower for trimethoprim-sulfamethoxazole-susceptible isolates (n = 670; MIC(90), 0.06 microg/ml) than for trimethoprim-sulfameth...

Streptococcus pneumoniae isolates resistant to several antimicrobial agent classes including trimethoprim-sulfamethoxazole have been reported with increasing frequency throughout the world. The MICs of trimethoprim, sulfamethoxazole, and trimethoprim-sulfamethoxazole (1:19) for 259 clinical isolates from South Africa were determined, and 166 of these 259 (64%) isolates were resistant to trimeth...

VIDENCE for the occurrence of mutation in the dihydrofolate reductase E gene of Diplococcus pneumoniae, which alters both the amount and structure of the corresponding enzyme, has been presented in two previous reports from this laboratory. (SIROTNAK and HACHTEL 1969; SIROTNAK, WILLIAMS and HACHTEL 1969). This form of dual biochemical effect is similar in some respects to that observed in the L...
